PH1894 | Zenker固定液 Zenker氏液 Zenker Fixative Solution （ZA固定液）

Zenker固定液是常用的混合型固定液，是细胞学及病理学常用的固定剂之一，由CG酸钾、S汞、乙酸和水混合而成。用该固定液固定的组织，细胞核及细胞浆染色十分清晰，特别是要显示免疫球蛋白、病毒包涵体(如Negri小体)、骨骼肌的横纹等组织时，应用本液可获得良好的结果。此液是病理解剖学、组织学和细胞学常用的固定液之一，显示细胞质的某些特殊颗粒有独特的优势，有利于胰岛组织细胞、垂体的一些细胞和心肌闰盘的显现。

注：Zenker 固定液工作液建议即配即用，溶液A也溶液B临用时混合。

产品特点：
1．Zenker固定液对细胞核固定效果较好，较适合于造血和网状内皮组织等样品的固定。
2．Zenker固定液不能用于保存细胞颗粒，红细胞及含铁血黄素。对含血量较多的组织标本不建议使用Zenker液固定。
3．常作为三色染色的组织固定剂，在三色染色中还作为媒染剂使用。
4．固定时间一般需要6-24小时。

储存条件：常温运输和保存，有效期一年。

使用方法：
1．配制适量体积的Zenker 固定液，按照19:1的比例将溶液A和溶液B混合，搅拌均匀，所得溶液即为Zenker 固定液工作液。
2．标本修块，置入Zenker 固定液工作液内固定6～24h(根据样品材料、大小不同，固定时间可能会有较大差异)。
3．一般1cm×1cm×0.2cm常规的组织块固定6h,流水冲洗1h转入低浓度乙醇开始脱水，染色前需用0.2%~0.5%的碘酒处理切片5~10min以除去汞盐沉淀，经水洗1min后用5%硫代硫酸钠或1%的硫酸钠水溶液处理切片2~5min至切片上的碘消失为止，再经自来水洗、去离子水洗后进行常规染


注意事项：
1．由于Zenker 固定液中含汞元素，固定后必须脱汞。
2．固定后，组织必须用流水冲洗去除CG酸钾否则乙醇脱水会引起络显色。

3．本固定液不能用金属容器盛放，组织固定后，也不能用金属镊夹取。

Zenker's fixative is a commonly used mixed fixative and one of the commonly used fixatives in cytology and pathology. The tissue fixed with this fixative has very clear staining of nuclei and cytoplasm, especially to show the inclusion of immunoglobulins and viruses. Good results can be obtained when applying this solution to tissues such as Negri corpuscles and skeletal muscle striated tissues. This solution is one of the commonly used fixatives in pathological anatomy, histology and cytology. It shows that some special particles in the cytoplasm have unique advantages, which are beneficial to the appearance of pancreatic islet tissue cells, some pituitary cells and myocardial intercalated discs.
